Abstract:

ABSTRACT: Objective   To explore the epithelial-mesenchymal transition phenomenon in silicotic nodule after SiO2 powders are douched as a one-off in rat trachea to make the silicotic model.

Methods   SiO2 powders (50mg/mL) and physiological sodium chloride (1.0mL) were douched as a one-off in rat trachea to make the silicotic model. The rats were divided into 4 groups: control 4w and 8w groups; silicosis 4w and 8w groups. Confocal laser scanning microscopy assay detected the colocalization of surfactant apoprotein A (SP-A) and α-smooth muscle actin (α-SMA) in the rat silicosis model. Immunocytochemical analysis determined the distributions of α-SMA and vimentin. The expressions of E-cad, SP-A, vimentin, α-SMA, type I collagen and collagen III were detected by Western blotting. Results   The silicotic nodule occured in rat lung and there was colocalization of α-SMA and SP-A in the rat silicosis model after SiO2 powders were douched as a one-off in rat trachea. As coincidence with the morphological changes of epithelial-mesenchymal transition, the expression levels of SP-A and E-cad decreased to 50% at silicotic 4w and to 21% and 35% at silicotic 8w respectively as compared with control groups. In contrast, silica increased the expressions of α-SMA and vimentin significantly by 1.6 folds and 3.7 folds at silicotic 4w, 1.6 folds and 2.9 folds at silicotic 8w compared with control groups, respectively. This process was accompanied by the increase in type Ⅰ collagen and collagen Ⅲ levels. The expression levels of type Ⅰ collagen and collagen Ⅲ increased to 1.7 folds and 1.6 folds at silicotic 4w, 1.6 folds and 1.8 folds at silicotic 8w, respectively, as compared with control groups (P<0.05). Conclusion   Silicosis fibrosis occurred in rat lung after SiO2 powders were douched as a one-off in rat trachea and epithelial-mesenchymal transition phenomenon also occurred in silicotic nodules.
