Abstract:
ABSTRACT: Objective   To investigate the association between polymorphisms of gastric cancer (GC)-related miR-21 target sites and the risk of GC. Methods   We enrolled 300 GC patients in Department of Gastroenterology and 300 healthy controls at Physical Examination Center of The First Affiliated Hospital of Xi’an Jiaotong University from January 2015 to December 2016. Blood samples were collected. The genotypes of miR-21 target sites, namely rs300574, rs4272, rs4728, rs11911, rs3744935, rs8708 and rs7828, were detected by MassARRAY method; the association between SNPs and risk of GC was evaluated by SPSS 19.0. Results   By comparing the allele frequencies of SNPs between the GC patients and controls using χ2 test, we found that the allele “C” of rs300574 in SPRY1 and the allele “T” of rs3744935 in Bcl-2 were associated with an increased risk of GC (P<0.05). In contrast, the allele “C” of rs8708 in JAG1 was associated with a decreased risk of GC (P<0.05). In the genetic model analysis, we found that under best model-dominant model, the T/C and C/C genotypes of rs300574 had a close correlation with an increased risk of GC (OR=2.64, 95% CI: 1.73-4.02, P<0.0001); the C/T and T/T genotypes of rs3744935 also exhibited an increased risk of GC (OR=1.58, 95% CI: 1.04-2.41, P=0.033). However, the C/T and C/C genotypes of rs8708 were associated with a decreased risk of GC (OR=0.42, 95% CI: 0.28-0.63, P<0.0001). The haplotype analysis results showed that the rs8708 and rs7828 in JAG1 were in strong linkage disequilibrium, and the haplotype “CA” constructed by rs8708 and rs7828 was significantly associated with a decreased risk of GC (OR=0.18, 95% CI: 0.13-0.44, P<0.0001).

Conclusion   The miR-21 target sites rs300574, rs3744935 and rs8708 may be associated with a decreased risk of GC.
